The tour begins from your hotel, where you will be picked up in an air-conditioned passenger van and shuttled deep into the back of Manoa Valley, seeing points of interest along the way (President Obama's childhood neighborhood, for instance).
The trail itself is an easy 20-30 minute hike through lush tropical rain forest. The trail passes mountain streams and pools, ancient Hawaiian lo'i kalo (taro patches), ohi'a 'ai(mountain apples), wild guava, mango trees, and even coffee trees.
Before long you will arrive at Manoa Falls, a 150-foot waterfall that has been the site for such shows as Lost, Hawaii Five-O, Jurassic Park, and more.
Go for the guided tour with a certified naturalist knowledgeable in everything you will encounter during your hike and then some. Two times available for the guided tour: 8am and 2pm. The guided tour involves about two hours of walking time, and also includes the use of a hiking stick.

The Manoa Falls trail is a relatively moderate trail with a slight incline and muddy (rainforest) terrain.
It takes most people 30minutes to one hour to hike to the waterfall, and usually a little less hiking back.
Bring water and bug repellant.
Wear comfortable, sturdy shoes that you don't mind getting muddy.
